Acess denied Very Big Trouble

I have a 500gb back-up external drive that Vista had locked me out of.
I have administrative access and I can edit the access to the drive (I have ownership and full access to this drive)
When I try to accss this drive it says "access denied"
I don't know how this has happened but is obviously part of Vista's accesss control.
Please Please Help Anybody

I can still access this drive (Thank God) on a different Computer but I need to use it on the one with Vista installed.

Yes I've done all that but It still tries to read the drive contents then stops and says access denied.

I guess you've turned off UAC also just to see if that is the problem. If so maybe someone else can pin point the problem. If taking ownership didn't work then I don't get what the problem is.
